The effect of fermentation time and addition of CMC to decrease of lignin content in fermentation of patchouli leaves using Phanerochaete chrysosporium

Abstract

Lignin is a compound that can obstruct the process of degradation of cell wall polysaccharides by a microbe. Cellulose can only be found in the form of binding to lignin and hemicellulose as lignocellulose. Cellulose and hemicellulose in lignocellulosic cannot be hydrolyzed by the cellulase and hemicellulase enzyme if lignocellulosic still contain lignin. The purpose of this study was to determine the effect of CMC and the fermentation time to decrease the lignin content and to determine the best operating conditions of the fermentation process patchouli leaves. The study was conducted with the following stages: drying and size reduction of patchouli leaf, up to 42 mesh, the regeneration of Phanerochaete chrysosporium in liquid media, patchouli leaf fermentation using a stirred bioreactor. Then analyzed lignin content before and after fermentation. Variable experiment was the addition of CMC (0, 0.4, and 0.8%), and time of fermentation (9, 11, 13, 15, 17, 19, and 21 days). The results showed that the fermentation time and the addition of CMC affect lignin content reduction. The favorable conditions: fermentation time of 21 days and the addition of 0.8% CMC, obtained lignin content of 12.238% and % reduction in lignin content 69.22%. © 2017 American Scientific Publishers All rights reserved.
